AnaSpec Launches Industry’s First Assay for Measuring Matriptase Activity

AnaSpec is pleased to announce the launch of the first commercially available kit for measuring the activity of Matriptase, a protease implicated in carcinogenesis.

The SensoLyte® Rh110 Matriptase Assay Kit provides a convenient assay for screening of Matriptase inhibitors and activators or for continuous assay of Matriptase activity using a highly sensitive fluorogenic substrate. Upon Matriptase cleavage, this substrate generates the Rh110 (Rhodamine 110) fluorophore which has a bright green fluorescence and can be detected at excitation/emission=496 nm/520 nm. The longer wavelength spectra and higher extinction coefficient of Rh110 provide greater sensitivity and less interference from other reaction components. This assay can detect as low as 0.15 ng/mL active Matriptase.

Matriptase (also known as MT-SP1, ST14, TADG-15 and epithin) is a trypsin-like protease from the family of type II transmembrane serine proteases. The spectrum of known Matriptase substrates includes extracellular matrix proteins, cell adhesion molecules, ion channels, growth-factor-like proteins and other proteases. Its actions can result in protein processing, activation or degradation. Matriptase is widely expressed in virtually all epithelium and is specifically found in tumors of epithelial origin. Matriptase has been implicated in carcinogenesis including ovarian, prostate and cervical cancers and is considered a drug target for developing cancer therapeutic agent (1-3).

HiLyte™ Your Antibodies Using One Universal Reagent

AnaSpec  is pleased to announce the release of a product line of recombinant Protein A, conjugated with HiLyte™ Fluor dyes or Biotin. This universal reagent can be used to detect antibodies from different species, such as human, rabbit, mouse, rat and others. Protein A is available as labeled with HiLyte™ Fluor 488 (Ex/Em = 499/523 nm), HiLyte™ Fluor 555 (Ex/Em = 553/568 nm), HiLyte™ Fluor 647 (Ex/Em = 649/674 nm), HiLyte™ Fluor 680 (Ex/Em = 678/699 nm), and HiLyte™ Fluor 750 (Ex/Em = 754/778 nm), or biotin. The large panel of emission wavelength HiLyte™ labeled Protein A enables the visualization of different target proteins within one species. Additionally, HiLyte™ Fluor dyes exhibit brighter signal compared to classic dyes (FITC, FAM, TAMRA, etc.).

Protein A is a non-glycosylated cell wall protein of Staphylococcus aureus that can bind the Fc part of immunoglobulin molecule of different species with strong affinity (1-4). Protein A consists of five IgG binding domains (A, B, C, D, and E), each approximately 60 amino acids long with no cysteines present and two S. aureus cell membrane binding domains – X and M (3).

AnaSpec’s recombinant Protein A consists of only IgG binding domains and was expressed in E.coli. Its apparent molecular weight is approximately 30,000 Da compared to the most reported 42,000 Da for the native Protein A that is found in S. aureus.

Recombinant Protein A conjugated with HiLyte™ Fluor 488 can be used for immunoglobulin detection in IHC and IF and for assay development.  Biotinylated Protein A can be used in the aforementioned applications, and for western blotting.

AnaSpec Introduces Two (2) Anti-Tau Antibody (GlcNAc Ser400 & Ser400) & Five (5) Tau Peptides

AnaSpec is pleased to announce the recent release of two rabbit polyclonal antibodies. Anti-Tau antibody (GlcNAc Ser 400) recognizes the glycosylated [N-acetyl-D-glucosamine (GlcNAc)] Serine400 residue. This antibody was raised against a synthetic peptide derived from amino acids surrounding the Tau Ser400 sequence. As confirmed by Western and dot blots, this antibody only recognizes Tau glycosylated Ser400 and not the non-glycosylated Tau. Species reactivity includes human, monkey, mouse, rat, and other mammalian species. As well, Anti-Tau antibody (Ser 400) that recognizes long variants of Tau (1N4R, 2N3R, and 2N4R) in human, monkey, mouse, rat, and other mammalian species was also introduced. The specificity of this antibody was confirmed by Western blot. This antibody recognizes all Tau modifications including phosphorylation and glycosylation.

In addition to the new antibodies, five new Tau peptides were also released. These peptides, 29 to 32 amino acids long are from Inserts1,  2 (from N-terminus) or the Repeat domains (C-terminus) of Tau isoforms found in human brain.

Tau is a microtubule-associated protein that stabilizes and promotes microtubule assembly. In the human brain, it is present in six isoforms that are 352, 381, 383, 410, 412, and 441 amino acids in length (1-5). When Tau is hyperphosphorylated, it can detach and form paired helical filament (PHF) aggregates (2-5). PHFs were found in neurofibrillary tangles in Alzheimer’s disease (AD) patients. As such, hyperphosphorylation of Tau has been linked to AD. In addition to being modified with phosphate groups, Tau can also undergo glycosylation on the same regions where phosphorylation takes place (5). Discovery of this phenomenon has led to the hypothesis that glycosylation of Tau may help stabilize microtubule-Tau connection and prevent free Tau aggregation.

AnaSpec Introduces SensoLyte® Deubiquitination Assay Kit

AnaSpec, EGT Group, is pleased to introduce the SensoLyte® 440 Deubiquitination Assay Kit. This kit provides a convenient assay for inhibitors/activators screening in deubiquitinylation or for continuous assay of enzyme activity. A natural substrate, ubiquitin, coupled with the AMC (7-amino-4-methylcoumarin) fluorophore is used in the kit. Upon cleavage by a deubiquitin protease, this substrate generates a blue fluorophore that can be detected at excitation/emission=354/442 nm. The substrate supplied in the kit is exquisitely sensitive for ubiquitin hydrolases (UCHs). The detection limit can reach as low as 3.7 pg/mL.

Modification of proteins with ubiquitin is a key step in protein degradation controlling many intracellular processes such as transcription, cell cycle progression, receptor internalization, and DNA repair.1 Recent studies have demonstrated that regulation also occurs at the level of deubiquitination.2,3 Deubiquitinating enzymes (DUBs) are proteases that reverse ubiquitin modifications.

Over 100 DUBs have been identified in humans. They are grouped into five distinct families based on their sequence similarities and mechanisms of action. Four of the families are cysteine proteases, while the fifth is a novel type of zinc-dependent metalloprotease.4,5 The majority of human DUBs belong to two cysteine proteases families, the ubiquitin specific proteases (USPs) and the ubiquitin carboxyterminal hydrolases (UCHs). Both protease families hydrolyze the peptide bond downstream of the C terminus of ubiquitin, which is either a classic peptide bond in the proforms of ubiquitin or an isopeptide bond to an e-amino group of a lysine residue within an ubiquitin modified protein. DUBs have been implicated in the pathogenesis of many human diseases, such as neurodegenerative disorders and cancer.6,7 Consequently they have become actively studied molecular targets for drug discovery.8,9

This kit, which employs a peptide substrate conjugated to a green fluorophore, is also good for inhibitors/activators screening in deubiquitinylation or for continuous assay of enzyme activity. Bright green fluorescence is generated upon protease cleavage and can be detected at excitation/emission=490/520 nm. The long wavelength spectra of the green fluorophore released provides less interference from other reaction components.

AnaSpec Offers Industry’s First Anti-MOG ELISA assay kits

AnaSpec is pleased to be the first in the industry to provide a whole suite of reagents and assay kits for the advancement of MS research. This includes MOG proteins, peptides, antibodies and assay kits.

Myelin Oligodendrocyte Glycoprotein (MOG), a protein with a molecular weight of 26- to 28-kDa, is a member of the immunoglobulin superfamily. It is expressed exclusively in the central nervous system (CNS), specifically on the surface of myelinating oligodendrocytes and external lamellae of myelin sheath. MOG protein constitutes only 0.01-0.05% of the CNS myelin proteins. However, it has been demonstrated to be a crucial autoantigen for multiple sclerosis (MS) in humans and experimental autoimmune encephalomyelitis (EAE) in rodents and monkeys (1-6).

These proteins, with sequences corresponding to human, mouse and rat extracellular domain, together with a 6x His tag were expressed in E. coli and purified from urea denatured bacterial lysate. The molecular weight found in all 3 recombinant proteins was 14.23 kDa. These proteins were shown to induce EAE in either mouse (for the human and mouse rMOGs) or rat (for rat rMOG).

AnaSpec Provides Industry’s Largest Collection of Anti-Tau Antibodies

In Alzheimer’s disease (AD), the major pathological hallmarks are β-sheet aggregates of β-amyloid peptides in senile plaques and hyperphosphorylated tau protein in neurofibrillary tangles (NFT).1-2  AnaSpec, a provider of one of the world’s largest collections of β-amyloid peptides, also provides the industry’s largest collection of anti-Tau antibodies – 23 phosphospecific and 14 non-phosphospecific Tau antibodies. These Tau rabbit polyclonal antibodies, raised against Tau peptides, are supplied as epitope-affinity purified rabbit IgG at 25 or 50 μg sizes at 0.2 mg/ml of 1x PBS solution. They have been validated in dot blots and IHC (Figure 1). Species reactivity includes human, mouse and rat. Some antibodies have also shown reactivity with bovine, chicken and zebrafish. Tau is a collection of microtubule-associated proteins that is involved in microtubule assembly and stabilization.3 In adult human brain, 6 isoforms, ranging between 352 and 441 amino acids in length, are produced as a result of alternative RNA splicing.4-5 The expression of Tau isoforms is developmentally regulated, as only the smallest Tau polypeptide is expressed in the fetal brain. Hyperphosphorylated Tau is the major component of the paired helical filament of Alzheimer’s disease. Anti-phospho-Tau antibodies are used to identify specific amino acids that are phosphorylated in Tau from normal brains and Alzheimer’s disease brains. The Tau proteins, especially in developing brains and in Alzheimer brains, are phosphorylated in vivo at many different sites such as Thr181, Ser198, Ser199, Ser202, Thr205, Thr212, Ser214, Thr217, Thr231, Ser235, Ser262, Ser356, Ser396, Ser400, Ser404 and Ser413.6-7
